The effect of miR145, miR155 on SKI expression in Namalawa cells in lymphoblastic leukemia

چکیده
Acute lymphoblastic leukemia (ALL) is a type of cancer that arises from the malignant transformation of lymphoblasts, the precursors of lymphocytes, in the bone marrow. In the context of ALL, various genetic and epigenetic changes contribute to the pathogenesis of the disease. Two vital genes involved in this process are SKI gene and TP53 gene. The SKI gene encodes a transcriptional repressor that plays a pivotal role in regulating gene expression related to cell proliferation and differentiation. In ALL, over-expression of the SKI gene results in suppression of tumor suppressor genes, inhibition of natural apoptotic pathways, and promotion of uncontrolled cell growth. Aberrant regulation of the SKI gene contributes to the pathophysiology of ALL by allowing malignant lymphoblasts to escape cell apoptosis. It also inhibits the TP53 gene. Research has shown that miR-145 can reduce the expression of the SKI gene and lead to a decrease in cell proliferation and an increase in apoptosis in ALL cells. The aim of the study is to investigate the effect of miR-145 and miR-155 on cell proliferation and apoptosis in acute lymphoblastic leukemia. Materials and methods: In order to investigate the effect, first NAMALWA cell lines related to lymphoblastic leukemia were obtained from Pasteur Institute of Iran and cultured in RPMI mediumnto reach the desired density in terms of number. Cells with optimal amounts of mimic sequences for miRNA-145 and miRNA-155, and cell proliferation and viability will be evaluated and extracted. Also, by qRT-PCR test, the expression level of SKI gene, which plays a role in cell proliferation and apoptosis, will be evaluated. GraphPad prism program, t-test and ANOVA tests will also be used for statistical comparison between different groups. Result: Our study showed that mi-145 has a significant increase compared to miR-155 in reducing SKI gene expression. In general, mir-145 inhibits SKI gene more than miR-155. The Findings: Our study showed that mi-145 has a significant increase compared to miR-155 in reducing SKI gene expression, and in general, mir-145 inhibits SKI gene more than miR-155. The p value of the results was <0.001.
