Evaluation of Anti-Proliferative Activity of Quercetin on lymphoma (Burkitt’s lymphoma) Cancer Cells and its effect on important microRNAs expressions specially miR451

Introduction:Quercetin is a plant-derived chemical found to have anticancer properties. Lymphomas are the most frequent haematological malignancy, putting a significant financial strain on the health-care system. Aim: In this study we evaluated the anti-proliferative action of quercetin on Burkitt's lymphoma cancer cells, as well as its effect on the expression of key microRNAs especially mir-451. Methods:Vitality of Raji cells in different concentrations of quercetin was tested using the MTT assay, and half maximal inhibitory concentration (IC50) was determined. MiR-451, -181, -155, -133, and -21 were then isolated. Each miR's primers and cDNA were created, and the expression level of each miR was measured using real-time RT PCR. Results: The amount of cell viability in the quercetin group is much lower and drops further as concentration increases. The IC50 was 30 µM. Quercetin enhanced miR-451 expression. Both quercetin and taxol/quercetin groups had a negative influence on miR-155, miR-133, and miR-181 expression, while taxol/quercetin had a considerably greater effect. Furthermore, miR-21 expression was lower in the quercetin and quercetin/taxol groups than in the taxol group. Conclusion:The recent investigation determined that the IC50 is equal to 30 µM. In addition, miRs -155, -181, and -21 as oncogenes dropped in the quercetin group, whereas miR-451 increased as a tumour suppressor. However, contrary to our expectations, miR-133, a tumour suppressor, was reduced in the quercetin group. The findings of this study, in conjunction with previous investigations, can be viewed as a foundation for future animal and clinical studies, potentially providing a medicine for controlling, diagnosing, and predicting the prognosis of cancer patients, particularly lymphoma patients.
