Evaluation of application of adipose stem cells with injectable gel in repair of cranial bone defect of rat

چکیده
The present study evaluated the combinatory effect of human PRP (hPRP) and collagen I (Col I) gels on the osteogenic potential of ASCs. Materials and methods: in vitro study is divided into eight groups: ASCs, ASCs/CaP, ASCs/PRP, ASCs/PRP/CaP, ASCs/Col I, ASCs/Col I/CaP, ASCs/PRP/Col I, ASCs/PRP/Col I/CaP. Biocompatibility, osteoblast differentiation, and mineralization were assessed through the MTT method, alkaline phosphatase assay, von kossa method and staining of osteocalcin immunocytochemistry, respectively. In in vivo study, 8mm rat calvaria bone defects were randomly divided into 6 groups and treated with either only defect, ASCs, PRP/Col I, PRP/Col I/CaP, ASCs/PRP/Col I and ASCs/PRP/Col I/CaP. All rat calvaria removed 12 weak after surgery and were examined by Ct scan and histology. Results: The in vitro results indicated that combination of PRP, Col I gels and CaP increased osteogenic differentiation of ASCs. In vivo results showed that combination of PRP, Col I gels and CaP increased bone reparation (P< 0.05)
