Evaluation of antifungal effect of combinantion of Aloevara extract and honey on Candida Albicans as invitro study
چکیده
Oral candidiasis is an opportunistic fungal infection which leads to acute and chronic infection in the mouth and gastrointestinal tract. The main cause of Candida infections is "Albicans". According to studies, common antifungal drugs have some side effects and sometimes they failed. So the purpose of this research, comparison of the antifungal effect of aloe vera extract and honey on Candida albicans with nystatin in vitro. Oral candidiasis is an opportunistic fungal infection which leads to acute and chronic infection in the mouth and gastrointestinal tract. The main cause of Candida infections is "Albicans". According to studies, common antifungal drugs have some side effects and sometimes they failed. So the purpose of this research ,comparison of the antifungal effect of aloe vera extract and honey on Candida albicans with nystatin in vitro.
method and materials
In this laboratory study, to evaluate the antifungal effect and determination of MIC (Minimum Inhibitory Concentration) of Honey Aloe vera extract, the CLSI (Clinical and Laboratory Standard Institute) protocol was used. Standard strains of Candida albicans (PTCC5027) were purchased and cultured. In addition to this fungus, Enterocoeus fecalis MMH12, Escherichia coli 29212 and WHONL Acenitobacter baumannii were also tested. After initial cultivation of the strains in blood agar , Bacteria were also cultured in TSB medium, and Also incubated for 24 hours at 37 ° C. Preparation of culture media and dilution of aloe vera extract and honey was done by macrodilution method. In this method, 4 different concentrations of Aloe vera extract and honey were evaluated on all 4 pathogenic strains. 16 tubes in total and for each species we placed two positive control and negative control tubes (24 tubes in total). The MIC of nystatin and gentamicin were also identified in this protocol. Macro dilution method and Agar well diffusion and colony count method were also used to verify the results.
Results:
MIC of aloe vera extract and honey for Candida albicans was 300 mg/ml. This result was confirmed by the colony count method. The results of all three methods confirm the excellent inhibitory effect and microbial susceptibility of Gram-negative bacteria and E.fecalis to the aloe vera extract and honey. This effect on Candida albicans is less and its effect is further enhanced by increasing concentration. Therefore, this compound cannot be a good substitute for nystatin, but can be used as a supportive treatment in candidiasis infections.
Conclusion
Increasing drug resistance to antifungal agents makes it necessary to examine new drugs to combat pathogen fungus. Aloe vera extract and honey having advantages such as low price, availability, better taste than Nystatin and low side effects could be a good candidate to be recommended as antifungal agent against infections caused by Candida albicans after passing clinical trials.
