Preparation and Evaluation of effect Magnetic Nanoparticles Modified with Biodegradable Copolymer PCL-PEG containing Curcumin – Silibinin on Leptin Gene Experession in Lung Cancer

Abstract
Lung cancer is the most lethal invasive disease in women and men .Leptin is a derivative of the adipocytes tissue has an important role in lung cancer. Curcumin (CUR) and silibinin (SIL) are herbal compounds which have many anticancer properties .CUR-SLN loaded MNPs PCLPEG improved inhibitory effect on leptin gene expersion. Purpose The purpose of this study was to investigate the efficiency of pure CURSLN and MNPs PCL-PEG on leptin gene expersion in A549 lung cancer cell line . Methods and Materials In the current study,the triblock copolimer PCL–PEG–PCL MNPs successfully synthesized by the method of ring-opening polymerization and encapsulated on PCL-PEG copolymer ,dual agents curcumin and silibinin . After A549 cell culturing ,MTT assay used to evaluated cytotoxic effects of free CUR-SLN and MNPs CUR-SLN and IC50 was calculated .Then A549 cells treaded with different concentrations of free CUR-SLN and CUR-SLN loaded on MNPs. Finally , leptin gene expersion level was compared by Real-time PCR. Result Synthesis of MNPs PCL-PEG and drug loading on nanoparticles was confirmed with using SEM, 1HNMR, FT-IR and VSM methods . Recent work has shown that CUR-SLN-loaded MNPs modified with PCL-PEGPCL triblock copolymers have potent time- and dose-dependent antigrowth effects in an A549 cell line. Real time PCR showed that CUR-SLN loaded on MNPs in compare with free CUR-SLN the target gene expression levels were reduced by half. 103 Conclusion Given the important role that leptin gene in lung cancer and magnetic nano-particle effective role in targeted drug delivery, nanoparticles containing curcumin and Silibinin as effective plant compounds to inhibit leptin gene expersion can be appropriate option.
