Design and preparation of complex delivery system formed from pectin and chitosan for encapsulation of resveratrol

Aime: Resveratrol is a promising candidate for the prevention and treatment of colon cancer. The low solubility and oral bioavailability of resveratrol limits its application. To overcome these problems, this work has aimed to develop chitosan-pectin nanoparticles (NPs) for colon targeted delivery of resveratrol. Material and methods: The effects of pectin: ZnCl2:chitosan (PZnC) %w/v, pH and ionic strength of media, addition of Poly Ethylen Glycol (PEG), and application of ultasonication on colloidal stability and release behavior of resveratrol from NPs were examined by High Performance Liquid Chromatography (HPLC), zeta potential, particle size analyzer, Scanning Electron Microscopy (SEM), and Fourier Transform-Infrared (FTIR) methods. Results: The results showed that the Particle size and Zeta of PZnC NPs in the ratio of~10:1:3 %w/v were 399±18 nm, +25±1 mV, respectively. Addition of PEG to system as a solvent for resveratrol (10%w/v) noticeably decreased the size of NPs to 83±3 nm. More than 63±4.3 % of resveratrol was encapsulated into the developed NPs and minor of release was observed in a duration of one month using simulated juice model (pH = 4±0.5) as investigated by HPLC analysis of resveratrol. antioxidant activity of delivery system in presence of resveratrol was about 93±6%. NPs released 13±0.9 and 38±1.2% of resveratrol in the simulated gastric and intestinal fluids, respectively. The remaining resveratrol (49±2.1%) was released in simulated colon fluid in the presence of pectinase and pH 7.4. It was concluded that our newly designed colloidal nanoparticulated system can be introduced as a novel platform for successful colon delivery of different drugs and bioactive component such as resveratrol. Conclusion: About a third of the amount of resveratrol added to the nano colloidal system-eventually reaches the colon and this amount is more than the minimum amount necessary for the therapeutic effects of resveratrol (50 mM).
