تعیین سطح بیان ژن های Myd88&TLR4,TLR2 در منوسیت های خون محیطی بیماران مبتلا به ویتیلیگو

Abstract

Aim: The aim of this study is to assess the gene expression level of TLR2, TLR4 and MyD88 in peripheral blood monocytes of vitiligo patients. Material and Method: The peripheral blood monocyte of vitiligo patients and the control group were isolated by Ficoll method and the adhesion ability to plastic surface. Monocytes purity was determined by Flowcytometery. Cell lysaet of the isolated cells were prepared and the gene expression level of TLR2 and TLR4 were analyzed by Real time RT PCR and MyD88 expression were studied by western blotting. Results: The results of Real time PCR and western blotting have been shown that the expression level of TLR2 and TLR4 and MyD88 reduced in peripheral blood monocytes of vitiligo patients., هدف: هدف ما از این مطالعه تعیین سطح بیان ژنهای TLR2، TLR4 و Myd88 در منوسیت های خون محیطی بیماران مبتلا به ویتیلیگو بود.مواد و روش ها: منوسیت های خون محیطی بیماران مبتلا به ویتیلیگو و گروه کنترل، با استفاده از فایکول و خاصیت چسبندگی منوسیت ها به سطح فلاسک کشت سلولی، ایزوله و درصد خلوص این سلولها توسط تکنیک فلوسایتومتری تعیین شد. سپس از منوسیت های ایزوله شده لایزت سلولی تهیه گردید و میزان بیان ژنهای TLR2 و TLR4 توسط روش Real time RT-PCR و میزان بیان ژن Myd88 توسط روش Western Blotting مورد ارزیابی قرار گرفت.یافته ها: نتایج حاصل از آزمون Real-time PCR و ایمونوبلاتینگ، نشان دهنده ی کاهش سطح بیان ژنهای TLR2، TLR4 وMyd88 در منوسیت های خون محیطی بیماران مبتلا به ویتیلیگو است.
