اهمیت کلینیکوپاتولوژیک بیان ژن تلومراز در تشخیص کانسر معده در نمونه های حاصل از لاواژ معده به روش semi quantitative PCR

Abstract

Goal of study: assessment of relative expression of telomerase's mRNA with Real-Time PCR in lavage fluids obtained from healthy people and the patients suffering from gastric cancer Methods: The present descriptive/cross-sectional study was carried out in Imam Reza medical-research center in Tabriz- Iran. With refer to previous documents and studies, the minimum number of samples were calculated for the study thirty patients with primary adenocarcinoma and fifty healthy people were selected for this study. Selection of the healthy and unhealthy subjects of the study was based on absolute random sampling. Total mRNA was extracted from the samples and revers transcribed to cDNA. The levels of telomerase gene expression in gastric lavage fluid was measured and compared utilizing Real-Time PCR. Findings: We calculate base activity as mean activity in 50 normal subjects and then comparing it with activity level of samples from patients reveal that there was a meaningful discrepancy between the rate of the telomerase gene expression in the samples taken from the healthy subjects and the samples from unhealthy subjects. All 30 patient's samples show higher level of activity than normal subjects. The mean activity in patient group was 6.653 (1.226 to33.980) fold compared to normal subjects., اهداف مطالعه: تعیین سطح بیان ژن تلومراز در مایع حاصل از لاواژ معده افراد سالم و تعیین سطح بیان ژن تلومراز در مایع حاصل از لاواژ معده افراد مبتلا نسبت به افراد سالم روش انجام مطالعه: پایان نامه به صورت طرح توصیفی مقطعی طرح ریزی و طی مدت یک و نیم سال در مرکز تحقیقاتی درمانی امام رضای تبریز انجام گرفت. تعداد نمونه برابر با 30 فرد مبتلا به آدنو کارسینوم اولیه معده و 50 فرد سالم با رجوع به مطالعات قبلی محاسبه گردید. انتخاب افراد به صورت کاملا تصادفی انجام شد. سطح پایه فعالیت ژن برابر با میانگین بیان ژن تلومراز در 50 شخص سالم محاسبه و نمودار توزیع فعالیت در افراد بیمار نسبت به آن ترسیم شد.یافته ها: درهر30 مورد سطح بالایی از فعالیت ژن ثبت گردید که متوسط فعالیت نسبت به افراد سالم6/356 برابر بود
