Effect of cordycepine on expression pattern of inflammatory and anti-inflammatory genes in chronic myeloid leukemia cells (CML)
Abstract
Our understanding of the role of inflammation in myeloid malignancies continues to expand. Chronic stimulation of cytokines leads to HSC depletion and provides a selective advantage for tolerant mutated clonal stem cells. However, what needs to be seen is how different types of selective stresses induce the growth of mutant HSCs. Interventions that maintain HSC fitness by reducing inflammation can potentially prevent the onset and / or progression of myeloid malignancies. Targeting inflammation can be as valuable as targeting a malignant clone, at least for chronic myeloid diseases.
Material and Methods: K562 chronic myeloid leukemia cell line was cultured in the relevant culture medium and after treatment with cordycepin, RNA was extracted. Then cDNA was synthesized. Finally, changes in the expression of IL-1β, and TNF-α genes and their control miRNAs were determined by qRT-PCR.
Results: QRT-PCR test changes in the expression of IL-1β, and TNF-α genes a significant decrease was observed, in treatment with cordycepin.
