Proliferation of callus from Nicotiana tabacum and nanoliposomes encapsulation of their extracts and its efficacy assessment in skin rejuvenation in vivo studies volunteers

Abstract
Introduction: Agents such as sun radiation may cause skin aging and hyperpigmentation. Using plants in cosmetic treatments is popular as they have fewer side effects. Utilizing plant stem cells is a new approach to formulating cosmetic creams that are plant-based. Plant stem cells are rich in antioxidants, growth factors, and many other compounds, therefore they are an invaluable resource of plant-based active ingredients. Different reports showed that tobacco inflorescence powder and leaves organic extracts enriched in polyphenolic compounds which are, consequently, an opportunity for a more sustainable extraction of antioxidant ingredients for the cosmetic industry, as they could be included in gels, lotions, and creams.Goal: We aimed to Investigating the anti-aging effects of extract from Nicotiana tabacum stem cell culture loaded in nanoliposome on human skin.Methods: Nano-liposomes were prepared by a thin layer method and characterized for size, Polydispersity Index (PDI), zeta potential (ZP),SEM, entrapment efficiency (%EE) and loading capacity (%LC). The in-vivo experiments on 12 volunteers were conducted with the Nicotiana tabacum extraction on nano-liposome on the periorbital fold area and the solution form of extraction on the and nasolabial fold area symmetrically for 2 months, and the effect of these two formulations on reducing wrinkles were compared. Results: The developed Nano-liposomes showed proper characteristics for dermal delivery ofNicotiana tabacum callus extract such as mean particle sizes around 110nm and EE values of %86.9 respectively. In-vivo studies in humans showed the superiority against the controls in the collagen generation in the skin and skin surface smoothness. Conclusion: It was concluded that the nanoliposome formulation containing Nicotiana tabacum callus extract can be introduced for local administration of Nicotiana tabacum callus extract in the prevention or treatment of photoaging.
