Evaluation of long non coding RNA LINC01111and DUSP1 expression level in tumor samples and healthy margin of head and neck melanoma

Abstract
Today, cancer is known as the second cause of death after cardiovascular diseases. Melanoma is known as the most common deadly malignancy involving the skin. These heads, whose origin is considered to be mutated melanocyte cells, have spread in recent years. has shown higher. Various environmental factors such as smoking and exposure to ultraviolet rays, along with genetic factors, are known risk factors of this disease. Carrying out projects such as transcriptome showed that most of the human genome is transcribed into non-coding RNA, these genes play different roles in regulating gene expression and various cell activities, and also play an important role in growth. and the progress of various diseases are similar. Among these, the role of the non-coding gene LINC01111 and its relationship with the level of expression of DUSP1 (MAPK-1) in different types of cancers have been investigated and taken into consideration. Materials and Methods: In this research, 30 tumor tissue samples and 30 tumor margin samples from cancer patients with head and neck mucosal melanoma including 17 men and 13 women were examined. RNA was isolated from the target tissues using an extraction kit (trizol), cDNA synthesis was performed from the extracted RNA, then Real Time PCR was performed to express the target genes, and finally statistical analysis was performed using the software. Results According to the results of the study, the expression of DUSP1 (MAPK-1) in the tumor tissue was significantly reduced compared to the healthy marginal tissue, but the expression changes of LINC01111 in the tumor and margin samples did not have significant changes. On the other hand, DUSP1 expression was related to clinical indicators of patients such as metastasis and lymph node involvement. Also, the findings related to biomarker analysis showed that the expression of these genes did not have the ability to distinguish healthy tissue from the patient and they have no biomarker value.
