Evaluation of TPO(rs1126799) and IL17A (rs8193036) gene polymorphisms in individuals with anti-TPO-positive serum levels and healthy individuals with normal Anti-TPO: a case-control study

Many studies have shown that inflammation contributes to the development of autoimmune thyroid diseases (AITDs). The present study was conducted with the aim of investigating polymorphisms of TPO gene (rs1126799) and IL17A gene (Rs8193036) in Anti TPO positive and negative patients.
Methods: In this clinical trial study, 128 Anti-TPO positive patients (mean age 41.48 ± 9.58 years) were introduced by the laboratory. After blood sampling, serum levels of vitamin D and TSH were measured. Based on TSH levels, they were divided into case and control groups. Finally, after DNA extraction, polymorphisms of TPO gene (rs1126799) and IL17A gene (Rs8193036) were investigated using tetra-ARMs PCR technique.
Findings: The results of our study showed that the difference in the frequency of rs1126799 genotypes including TT and TC between the case and control groups was not significant (P>0.05). Regarding Rs8193036 genotypes, there was no significant frequency difference for TT and TC between case and control groups (P>0.05). Hardy-Weinberg equilibrium analysis of the rs1126799 locus for case individuals showed that the difference observed for the observed and expected genotypes between the two groups was not significant and the population was in equilibrium. But for the position Rs8193036, the difference observed for the observed and expected genotypes between the two groups is significant and the population is out of balance.
