Evaluation of VDR (rs7975232) and TPO (rs2071400) gene polymorphisms in individuals with anti-TPO-positive serum levels and healthy individuals with normal Anti-TPO

Many studies have shown that low vitamin D levels contribute to the development of autoimmune thyroid diseases (AITDs). This study was designed to investigate vitamin D levels and its relationship with anti-thyroid peroxidase (anti-TPO) antibodies in patients with autoimmune hypothyroidism. The present study was conducted with the aim of investigating polymorphisms of VDR gene (rs7975232) and TPO gene (rs2071400) in Anti TPO positive and negative patients. Methods: In this clinical trial study, 128 Anti-TPO positive patients (mean age 41.48 ± 9.58 years) were introduced by the laboratory and divided into hypothyroid and euthyroid based on TSH level. Then serum levels of vitamin D, TSH and Anti-TPO were measured. After blood sampling and DNA extraction, polymorphisms of VDR (rs7975232) and TPO gene (rs2071400) were investigated using tetra-ARMs PCR technique. Results: The difference in frequency of rs7975232 genotypes including AA, CA and CC between hypothyroid and euthyroid groups was not significant (P>0.05). Regarding Rs2071400 genotypes, there was a significant frequency difference for CC and CT between hypothyroid and euthyroid groups (P=0.028). This is if the TT genotype was not observed in any of the two groups. Also, the calculated OR is significant. As a result, CC genotype reduces the risk of disease and increases health, and CT genotype increases the risk of disease and decreases health. Hardy-Weinberg equilibrium examination of the rs7975232 and Rs2071400 loci for hypothyroid individuals showed that the difference observed for the observed and expected genotypes between the two groups was significant and the population was out of balance.
