Investigating the effects of knock-down of SlC16A13 on apoptosis and metastasis of thyroid cancers cell line

Abstract
The role of imbalaced gene expression is well known in thyroid cancers especially in medullary and non medullary types. The aim of this investment is to investigate the effect of knock-down of SlC16A13 on apoptosis and migration in the TPC 1 cell line of human papillary thyroid cell line. Materials and methods: Tpc 1 cell line, representative of papillary thyroid cancer cells, were cultured in RPMI + 10% FBC medium. Then the synthetic anti SLC16A13 shRNA was transfected into the target cell line. Then the effect of transfection on the expression change of its target was approved by Real Time PCR. The effect of this treatment on the cell viability and apoptosis was evaluated by MTT and flow cytometry tests, respectively. The SCRATCH test was also used to investigate the role of this treatment on the ability of cancer cells to migrate. Expression of genes involved in apoptosis and cell migration including Casp-3, Casp-8, Casp-9, Bcl2, Bax, E-cadherin and Vimentin also have investigated by qRT-PCR. Results: We observed that knocking down of SLC16A13 in the TPC 1 cell line of papillary thyroid cancer led to an increase in the rate of apoptosis but does not effect on the abillity of the cells migration potential. Also, the examination of the expression of genes involved in the process of apoptosis and migration showed that the expression of Casp-3, Casp-8, Casp-9, Bax, Vimentin genes had increased, and the changes in the expression of Casp-3, Casp-8, Bax were statistically significant. Another gene investigated in the present study was BCL-2, which showed significantly decrease in expression rate. In this study, the expression level of E-CADHERIN gene was also analyzed, and the changes of this gene were not statistically significant.
