Evaluation of expression of the relationship between the expression of UCA1 lncRNA and miR-143 genes in tumor tissue and tumor margin tissue in patients with tongue SCC

Abstract
SCC of the tongue is one of the most common cancers related to the oral cavity. This disease has a very complex pathogenesis, and researches so far have not been able to determine the main reason and how this disease progresses, this shows the importance of investigating the expression profile of genes in this disease to reveal the molecular process of the pathogenesis of this disease. In this direction and to improve the understanding of the molecular pathways that cause SCC of the tongue, this study was conducted with the aim of determining the expression profile of UCA1 lncRNA and the expression level of miR-143 in tumor and margin samples of SCC of the tongue. Methods: Tissue samples, including 50 tongue SCC cancer tissue samples and 50 margin tissue samples, were prepared and subjected to RNA extraction. Complementary DNA was synthesized and then the expression levels of target genes were measured using real time PCR. In addition, the relationship between the expression of the studied genes with each other and the clinical pathological characteristics of the patients was evaluated. Results: The results obtained from q-PCR showed that both studied genes had significant expression changes in tumor samples compared to the margin (p>0.05). The expression level of UCA1 lncRNA in tumor samples compared to healthy tissue had There was an increase, but the expression of miR-143 was decreased in the samples. Also, the changes of both genes were related to metastasis and involvement of lymph nodes. In addition, the expression of the two desired genes in the tumor samples had a negative correlation.
