Isolation ،characterization and cytotoxicity investigation of the exosomes secreted by Doxorubicine-loaded cyclodextrin nanosponge primed cells
Abstract
Introduction:Due to the increase in cancer worldwide, methods for effective treatment of cancer and prevention of disease progression have attracted the attention of scientists and researchers. Among these methods, the use of exosome alone or together with nanoparticles as a carrier.Aim: Isolation ,characterization and cytotoxicity investigation of the exosome secreted by doxorubicin-loaded cyclodextrin nanosponges primed cells,this is a useful method for cancer treatment .Methods: Here, we describe a technique for exosome purification from cell culture, and Synthesis of nanoparticles., and further analysis by SEM microscope, and DLS .Investigating of cytotoxicity effect of doxorubicin-loaded cyclodextrin nanosponge primed cells,investigating drug and doxorubicin –loaded nanosponge penetration inside the cells.Results: Examining the shape and size of nanoparticles with SEM showed that the produced nanoparticles are spherical and their average size is 168-71 nm. It was observed by examining dried exosomes from macrophages treated with nanoparticles containing doxorubicin.The secreted exosomes did not have any effect in reducing cancer cell growth.Conclusion: The exosomes that were secreted from the media with nanoparticle containing drugs had no effect on the death of cancer cells Due to their special characteristics, exosomes have had a great impact on cancer treatment in the past few decades, and the shape, size, and surface characteristics of nanoparticles have an effect on the proliferation or destruction of cancer cells.
