Evaluation of the effect of MiR-206 on SKI gene expression by intervening NF-KB pathway in A-375 melanoma cell line

Abstract
Introduction: Skin cancer is one of the most common cancers in the world. One out of every three cancers diagnosed in the world is skin cancer, and the importance of early treatment is increasing day by day. Melanoma is the most aggressive and deadly type of skin cancer and one of the most malignant cancers. Various methods such as radiotherapy, chemotherapy, immunotherapy and surgery are used in the treatment of this type of cancer. Materials and methods: The present laboratory study was performed on melanoma cancer cells in RPMI640 culture medium containing 10% FCS at 37ₒc and 5% CO2. After counting and dividing the cells, the inside of the cell plate was stimulated with MiR206. Incubation was done. After 48 hours, the cells were collected and total mRNA was extracted and the expression level of SKI was checked through qRT-PCR. Statistical tests P < 0.05 were considered significant. Results: The results of the present study showed that the expression level of SKI in melanoma cell line (A-375) decreased. Also, the results showed the level of NF-KB expression in melanoma cell line were decrease.
