Evaluation of the effect of MiR-206 on the expression of TLR2, TLR4 in melanoma A-375 cell
Abstract
Cancer is a group of diseases whose main feature is unregulated cell growth, invasion and spread of cells from the original location to other parts of the body. Despite recent advances in diagnosis and treatment, cancer is one of the leading causes of death in the world. Skin cancer is one of the most common cancers in the world. The aim of this study was to determine the effect of MIR-206 on TLR2 and TLR4 in melanoma cell line (A-375).
Materials and Methods:
The present in vitro study was performed on melanoma cancer cells in RPMI640 culture medium containing 10% FCS at 37 ° C and 5% CO2. After counting and dividing the cells, the cells inside the plate were stimulated in a time course with MIR-206 protein. Incubation was performed. After 48 hours, the cells were collected and total mRNA was extracted and the expression levels of TLR2 and TLR4 were examined by qRT-PCR. Data were analyzed using t-test and SPSS software, and P<0.05 was considered significant in all statistical tests.
Results:
The results of the present study showed that the expression of TLR2 and TLR4 decreased in melanoma cell line (A-375) in exposure to MIR-206.MTT results of melanoma cell death were compared at different times and statistical analysis showed a significant difference (P<005).
