Evaluation of SIK1-LNC and SIK1 expression patterns in tumor tissue and free margin in patients with lung cancer

Abstract
Lung cancer is the second most common cancer and the main cause of cancer-related death among women and men all over the world. In the present study, we investigated the expression pattern of SIK1 and SIK1-LNC in tumor tissue and tissue with healthy margins in lung cancer patients. Materials and methods: In this study, 100 tissue samples (50 tumoral samples and 50 samples with healthy margins) were used, which were collected during previous examinations and were transferred to RNA storage solution by the surgeon. . Total RNA was extracted using the mentioned samples and with the Qiagene kit. Then, using the PolyA RT-PCR method, all the mRNAs in the sample were converted into cDNA, and after designing the primers, they first checked the expression of the target genes using the PCR machine, and after confirmation, using the Real- time PCR, we investigated quantitatively the expression level of the target genes in the examined samples. Results: In this study, 50 samples from male lung cancer patients and 50 samples from the healthy margin of these tissues were examined. Of these, 29 people (58%) were over 55 years old and 21 people (42%) were under 55 years old. Most of the examined patients were in stage 3 of the disease (48%). Examining the patients for the presence of lymphatic metastasis has shown a higher frequency of patients with metastasis (29 vs. 21) and also most of the patients mentioned a history of receiving chemotherapy (40 cases, 80%). Also, the level of gene expression in samples taken from patients with lymph node involvement and patients who did not receive any chemotherapy was higher than their opposite group (both cases P < 0.05). The results of roc analysis are the results of the relevant analysis showed that this gene, separately from lymph node involvement, receiving chemotherapy status and general analysis, in all three cases, the area under the graph is 0.71% (P=0.0027). The expression level of SIK1-LNC gene has decreased significantly in tumoral tissue compared to healthy peripheral tissue (P<0.05). Examining the correlation between the expression of SIK1-LNC and SIK-1 genes showed a significant positive correlation (r=0.4, P=0.007).
