The effect of PDL-1 silencing on Kidney carcinoma cell survival

Abstract
Kidney cancer is actually a cancer that starts in the kidneys. Your kidneys are two bean-shaped organs, each about the size of your fist. They are located behind the abdominal organs, so there is one per spine. Renal cell carcinoma is the most common type of kidney cancer in adults. Young children are most likely to get a type of kidney cancer called Wilms' Tumor. High expression of PDL-1 by cancer cells is known. Since the effects of antibodies against PD1-PDL-1 are known, alternative treatment methods such as siRNA can be used to suppress a specific gene. In this study, the aim is to suppress the expression of PDL-1 genes by using specific siRNA. and evaluate the effect of this simultaneous suppression on the growth, migration and apoptosis of cancer cells. Materials and methods: First, the cell line (ACHN) was cultured. Then the specific siRNA of PDL-1 gene was transfected into the desired cell line. MTT test was performed to check the cytotoxic effect of transfected siRNA. In order to investigate this gene suppression on the rate of growth and proliferation of cancer cells, genes involved in growth, proliferation and metastasis (cMyc, Ki67, BCL-2, MMP9) were investigated by PCR-RT method. After collecting data in Excell software, data organization was done and then data distribution was checked using Prism software and for normal distribution by Test-T test and for data with non-normal distribution by Whitney test. -Mann was used. Results: We observed a significant increase in PDL-1 expression in kidney cancer cell line. Compared to the control group, our results showed a significantly decreased frequency of genes involved in proliferation and cell migration such as Ki67, MMP9, BCL-2 and C-myc after adding PDL-1 siRNA. Also, the data obtained from MTT showed that after adding PDL-1 siRNA, the rate of growth and proliferation of cancer cells decreased.
