Evaluation of gene TMPO-AS1 expression s in tumor tissue and marginal tumor of thyroid cancer patients
Abstract
The role of long non-coding RNA molecules (lncRNAs) in thyroid cancer is well known. The purpose of this study was to investigate the expression changes of TMPO-AS1 gene as an important lncRNA in the process of tumorigenesis in tumor tissue and tumor margins of thyroid cancer patients.
Method: This research was a case-control study and the target population was patients with thyroid cancer in the northwest of the country in 2021. The final sample size was 50 people. After receiving permission from the Ethics Committee of Tabriz University of Medical Sciences, 2 tissue samples from the tumor and margin were taken from the patients during surgery and immediately transferred to the preservative solution. In order to check the expression of genes, RNA was extracted using Trizol and then synthesized by cDNA synthesis kit and then the expression level of the desired genes was checked using Real Time PCR method.
Results: In this study, TMPO-AS1 gene expression in thyroid cancer tissues increased significantly compared to adjacent tissues (P<0.05). ROC curve analysis for TMPO-AS1 gene showed that this gene had a good ability to diagnose patients with thyroid cancer due to the level under the graph equal to 0.91. Also, TMPO-AS1 expression had no significant relationship with age, smoking, family history, lymph node metastasis, metastasis and gender of patients. But there was a significant relationship with the disease stage variables (P=0.023) and capsular involvement (P=0.043).
