Study of the effect of cordycepine on the expression of immune checkpoint genes in gastric cancer cells

Abstract
Introduction: timely diagnosis and correct and targeted treatment of gastric cancer are of great importance. One of the important factors in tumor spread is the expression of immune checkpoint molecules, inhibition and escape from the immune system. miRNAs are a group of non-coding RNAs with a sequence of about 22 nucleotides that lead to the regulation of gene expression. Regulating immune check points provides a breakthrough in cancer treatment. Blocking this novel immune check point can enhance the efficacy of cancer treatment. Methods: In the current study, at the post-transcriptional level of MKN-45 cell cultures in RPMI1640 culture medium and after reaching logarithmic growth of cells, the two groups They will be treated with Cordycepin and treated with the standard drug Docetaxel. After 24 hours, the cells will be collected for mRNA and miRNA extraction. To normalize the results, U6 will be used to examine the differences in miRNA expression and beta-actin to examine the differences in mRNA expression in the Real-time PCR test. Results: We observed a decrease in the expression of immune checkpoints PD-L1 and CD80 gene after treatment with cordycepin. And also the expression of miR155 and 424, that regulates immune checkpoints, had been increased.
