Evaluating the expression levels of lncRNA DLGAP1-AS2 in colorectal cancer tissues as a diagnostic biomarker
Abstract
Colorectal cancer is one of the most common cancers worldwide. The role of lncRNAs in the development of various types of cancer, especially colorectal cancer, has been reported. Therefore, in this study, the expression level of lncRNA DLGAP1-AS2 was investigated in colorectal cancer and adjacent healthy tissues.
Methods: First, the expression of DLGAP1-AS2 lncRNA was evaluated using TCGA database in colorectal cancer samples compared to healthy samples. Then the number of 33 colorectal cancer tissues and 33 adjacent healthy tissue samples were taken from colorectal cancer patients with written consent and their clinical and pathological characteristics were determined and determined by the pathologist. Then RNA related to the samples was extracted and after checking and confirming their quality and concentration, cDNA synthesis was performed. In the following, the expression of lncRNA DLGAP1-AS2 in the samples was measured by real-time PCR method and its relationship with clinical and pathological characteristics was investigated. Finally, statistical analyses were performed to determine the biomarker power of this lncRNA.
Results: The results of TCGA datasets and laboratory samples showed that the expression level of lncRNA DLGAP1-AS2 in colorectal cancer samples increased significantly compared to healthy adjacent samples. Also, the results of ROC curve analysis in TCGA samples indicated the biomarker power of this lncRNA.
