Prepration and Evaluation of Physicochemical Characteristic of Co-Q10 Immediate release pellets
Abstract
Introduction: Many of new discovered drugs have poor water solubility.this problem causes low oral bioavailability. One solution to improve the bioavailability is to prepare self-micro emulsifying drug delivery system (SMEDDS).This isotropic system formed fine O/W emulsions under genyle agitations of gastrointestinal movements . to overcome disadvantages of liquid SEDDS , this form could be transformed to solid dosage forms.Aim: the Aim of this study is formulation and evaluation of solid self-microemulsifying immediate- release pellets of CO-Q10
Methods: for identifying the best components of liquid SMEDDS,solublality of CO-Q10 carried out in different oils ,L-seeds prepared in different ratios of oil,surfactant and cosurfactant.then self-micro emulsifying formulations were formed and by using of Avicel,PVPK30 and superdesintegrant and they were turned to wet mass and yellow-colored pelletd were formed using Extrusion-Spheronazation machine. these pellets were evaluated of dissolution rate,size distribution,load of drug in formulation,bulk and tapped density and siza and shape charactristics.
Results: CO-Q10 has the most solubility in Migliol as oil,Myrj 52 and Cremophor EL as surfactant and Transcutol P as cosurfactant so they were used as components of formulations .11 formulations were made and further evaluations shows that size distribiution of pellets were between 700-850 microns, density of pellets were between 670 to 750 g/l .all of the formulations had fair Flowability . dissoloution rate of self microemulsifying pellets were dramatically increased compared to formulation number 1 which weren’t made by self-microemulsifying formulation. According to USP pharmacopeia about immediate- release formulations ,formulations containing Cremophor EL as surfactant had dissoloution rate of more than 80% in 15 minutes.disintegration tests also demonstrated the importance of superdisintegration of in an immediate release formulations.
Conclusion: self microemulsifying immediate release pellet of CO-Q10 is useful way useful way of increasing solid form absorption of CO-Q10.
