MicroRNA-326 and microRNA-155 in recurrent miscarriage patients before and after lymphocyte therapy in comparison to control groups

Abstract
Background : Recurrent pregnancy loss (RPL) or recurrent spontaneous abortion is defined as at least two consecutive clinical pregnancy losses before 20th week of gestation. Paternal antigens, which are expressed by early embryo, cause maternal immune system to response, therefore, regulatory mechanism are required to make the immune response tolerate the foreign antigens. In this study, we will review the role of epigenetic factors and especially microRNAs in function of different T-cells. MicroRNAs (miRNAs) are short (approximately 22 nt) noncoding RNA molecules regulating expression of target genes at the posttranscriptional level by translational repression or messenger RNA degradation. Goal : we investigated MicroRNA-326 and microRNA-155 in recurrent miscarriage patients before and after lymphocyte therapy in comparison to control group.Methods : To prepare PBMCs, first 10 ml of heparinized blood is taken from patients and peripheral blood mononuclear cells are isolated using Ficol 1.077. According to the standard protocol, the number of 2 x 107 is injected subcutaneously to patients with recurrent miscarriage. The number of injections is three times with one month intervals. Real-time PCR technique will be used to check the expression level of microRNA-326 and microRNA-155 molecules. Findings : The gene expression level of micro-RNA 155 in RPL women before three cycles of subcutaneous injection of lymphocytes was 1.584 ± 0.5633 and the gene expression level of the same micro-RNA after the lymphocyte therapy of the mother was 204 ± 0.4350. / is 1. Also, the expression level of micro-RNA 326 gene in these women was reported to be 1.662±0.5799 before treatment and 1.662±0.5799 after three cycles of treatment with paternal lymphocytes. This level of gene expression in women with a history of successful pregnancy as a control group, for micro-RNA 155 and micro-RNA 326 genes is 1.000 ± 0.1021 and 1.000 ± 0.0659, respectively.Conclusion : A noticeable decrease in the gene expression of microRNAs under study
