Formulation of osmotic drug delivery system with porous membrane of empagliflozin and investigation of drug release from it
Abstract
Introduction: One of the benefits of osmotic pump is constant release rate, long release time and non-dependence of release factors on the body's digestive system. Among the oral osmotic pumps, the CPOP (controlled porosity osmotic pump) system has a very simple and highly effective structure. Aim: The purpose of preparing an osmotic pump with a porous membrane is to release uniformly and close to zero degree.Method: The central core was prepared by direct compression and the core was coated through dip coating. The membrane formulation in this system includes acetate (CA) as a semi-permeable membrane and castor oil as a lipophilic plasticizer and polymer. PEG is used as a hole-forming polymer. Cellulose acetate phthalate polymer was also used as an enteric coating in the formulation.Findings: The four parameters used to compare the prepared formulations include D24h ,tl (Drug release delay time from the system).
Discussion and conclusion: The results show that the thickness of the membrane was found to be half between the type and concentration of pore former and parameters. Different concentrations of PEG in semi-permeable membrane as a hydrophilic plasticizer significantly reduced the effect on tl and D24h. While increasing concentration of hydrophilic plasticizers decreased tl and increased D24h.
