Evaluation P14 and P16 expression in tumoral and marginal tissues of patients with thyroid cancer
Abstract
The role of genetic factors in thyroid cancers is well known, especially in medullary and non-medullary familial types. The aim of this study was to evaluate P14 and P16 expression in tumoral and marginal tissues patients with thyroid cancer.
Materials and Methods: This research was a case-control study and the target population was patients with thyroid cancer in the northwest of the country in 1400. The final sample size was 75 people. After receiving permission from the ethics committee of Tabriz University of Medical Sciences, 2 tissue samples from the tumor and marginal tissues were taken from the patients during surgery and immediately transferred to the preservative solution. After that, the samples were transferred to the immunology research center of Tabriz University of Medical Sciences, following the cold chain, for further investigations. In order to check the expression of genes, RNA was extracted using Trizol and then synthesized by cDNA synthesis kit, and then the expression level of the desired genes was checked by using Real Time PCR method.
Results: In this study, the expression level of P16INK4A gene in thyroid cancer tissues was significantly increased compared to adjacent tissues (P-value=0.003). Meanwhile, the expression of P14ARF gene in tumor tissues showed a lower expression than the margin tissues (P-value=0.014). The analysis of the ROC curve for the P16INK4A gene showed that it had a level under the graph equal to 0.6620, which due to the statistical significance (P-value=0.004424) of this gene can be used as a therapeutic target or a criterion for The diagnosis of thyroid cancer patients should be studied further. ROC curve analysis for P14ARF gene, which has an area under the curve equal to 0.5782 due to the lack of statistical significance (P-value=0.169), it was determined that this gene, unlike the previous gene, does not have such a capability. In this study, among the demographic and background factors, the disease stage variables (P.value=0.004), Lymph node metastasis (P.value=0.001), Metastasis (P.value=0.001) and Capsular involvement (P.value=0.001) had a statistically significant relationship with P14ARF gene expression; On the other hand, other variables such as gender, age, family history and smoking had no statistically significant relationship (P.value>0.05). Also, among the demographic and background factors, the variables Lymph node metastasis (P.value=0.004) and Metastasis (P.value=0.002) had a statistically significant relationship with P16INK4A gene expression; On the other hand, other variables such as gender, age, disease stage, capsular involvement, family history and smoking had no statistically significant relationship (P.value>0.05).
