The effect of Forskolin and Metformin alone and in combination on sperm parameters, sexual hormones and testis histological alteration in type 2 diabetic rats
Abstract
Diabetes is a chronic disease that could cause damage to the reproductive system. This study aimed to investigate the effects of forskolin and metformin alone and together on spermatogenesis and blood biochemical factors in adult type 2 diabetic rats.
Materials and Methods: Animals were randomly divided into 5 groups. Control group, type 2 diabetes, type 2 diabetic treated with forskolin (100 mg/kg), type 2 diabetic treated with forskolin (300 mg/kg), type 2 diabetic treated with forskolin and metformin. After the treatment period (8 weeks), oxidative stress markers were measured and Bax and Bcl-2 expression were assessed by RT-qPCR. Sperm parameters and hormone levels were also examined.
Results: Johnsen’s score along with epithelial thickness and seminiferous tubule diameter increased in the diabetic group after the treatment with forskolin and metformin alone and together compared with the diabetic group. The testosterone, LH, FSH and antioxidant enzymes levels in the treatment groups increased significantly compared to the diabetic control group. MDA levels and Bax expression showed a significant decrease in the treatment groups. Bcl-2 expression was also increased in the treated groups.
