Evaluation of the sex hormones level and oocyte maturation associated genes in the cryopreserved ovary after transplantation in fibrin scaffold with endothelial cells and melatonin in rat
Abstract
In this experimental study, 36 adult female rats (8 weeks) underwent ovariectomy. Then their ovaries were divided into six groups: control, Vitrification/transplantation (V/T), V/T with fibrin scaffold (V/T-S), V/T with fibrin scaffold and melatonin (V/T-S-M), V/T with fibrin scaffold and endothelial cells (V/T-S-C), V/T with fibrin scaffold, endothelial cells, and melatonin (V/T-S-M-C). Finally, the ovaries autotransplantation in the rats' lumbar region. After 14 days, the ovaries were removed. The level of 17-beta estradiol and progesterone hormones was evaluated by the ELISA technique, and expressions of genes BMP-15 and GDF-9 were related to the maturation oocyte by real-time RT-PCRResults: This research showed that the expression of GDF-9 and BMP-15 genes and the level of 17β-estradiol and progesterone hormones decreased significantly in the transplanted frozen ovary group (P<0.05) and increased in encapsulated ovary groups. This increase in the expression of GDF-9 and BMP-15 genes and the level of 17β-estradiol and progesterone hormones in the ovaries encapsulated with fibrin scaffold and melatonin, and ovaries encapsulated with fibrin scaffold along with melatonin and endothelial cells is a significant difference with the transplanted frozen ovary group (P<0.05). The comparison between treated groups showed that the expression of GDF-9 and BMP-15 genes and the level of 17β-estradiol and progesterone hormones were higher in the groups containing melatonin than in the group containing fibrin scaffold and fibrin scaffold with endothelial cells. Also, no significant difference was observed between the ovaries encapsulated with fibrin scaffold and melatonin, and fibrin scaffol with melatonin and endothelial cells. The presence of endothelial cells had no significant effect on increasing the expression of GDF-9 and BMP-15 genes and the level of 17β-estradiol and progesterone hormones
