Abstract
Diabetes, as a debilitating disease, has caused an increase in mortality in human societies due to its negative consequences and various complications, including cardiovascular complications. The purpose of this study was to investigate the effect of intravenous injection of stem cells + c-kit on the expression of mitochondrial biogenesis genes (PGC-1a, SIRT1, Tfam) in the hearts of type 2 diabetic male rats.
method:
58 male Wistar rats in 4 control groups (n=12), diabetic group (n=12), diabetic group + c-kit+ cells (n=12) and diabetic group + c-kit ¯ cells (n=12) were randomly divided. The animals of the diabetic groups received a high-fat diet to become diabetic, and then they were injected with streptozotocin at a dose of 35 mg/kg, 50 microliters of normal saline solution to the diabetic group and 50 microliters of normal saline solution containing about three hundred thousand c-kit cells to the diabetic group + c-kit cells And 50 microliters of normal saline solution containing about three hundred thousand c-kit ¯cells were given intravenously to the diabetic group + c-kit¯ cells. Finally, 60 days later (causing cardiac changes), the animals were anesthetized with ketamine and xylazine, and immediately the heart (APEX) was removed for histological studies and the rest of the left ventricle to measure the expression level of PGC-1α, SIRT1, Tfam as indicators of mitochondrial biogenesis, and the results It was investigated and analyzed with the one-way ANOVA test.
Results:
The reported results of this study showed that the expression level of SIRT1 gene decreased in all diabetic groups compared to the control group, and this decrease was significant only in the diabetic group. But the values of this gene increased in the group receiving C-KIT+ compared to the diabetic group, which was not significant. Also, the results of the above study showed that the values of Tfam and PGC-1α genes decreased in diabetic groups (group D and C-KIT¯) and showed that this decrease was significant in PGC-1α gene but not significant in Tfam gene. Also, the values of these genes in the group receiving C-KIT+ showed a significant increase compared to the diabetic group. Also, histological study showed fibrotic changes in the left ventricle.
