The effect of intravenous injection of c-kit+ stem cells on ABCA1 gene expression levels and inflammatory factors in type 2 diabetic rats

Abstract
Diabetes as an inflammatory disease causes an increase in heart disorders and ultimately increases the death rate in people. The purpose of this study was to investigate the effect of injection of c-kit +⁺ stem cells extracted from bone marrow on ABCA1 gene expression and inflammatory factors in type 2 diabetic male rats. method: 58 male Wistar rats were randomly divided into 4 control groups (n=12), diabetic group (n=12), diabetic group + c-kit+ cells (n=12) and diabetic group + c-kit- cells (n=12). were divided. Diabetic groups were diagnosed with diabetes by receiving a high-fat diet and intraperitoneal injection of streptozotocin (35 mg/kg). The diabetic group received 50 microliters of normal saline solution and the diabetic group + c-kit+ cells, 50 microliters of normal saline solution containing about 300000 c-kit+ cells and the diabetic group + c-kit- cells, 50 microliters of normal saline solution containing about 300000 c-kit- cells intravenously they received. Finally, 60 days later (causing cardiac changes), the animals were anesthetized with ketamine and xylazine and immediately the heart (APEX) was removed for histological studies and the rest of the left ventricle to measure the expression level of ABCA1 and IL-10 and IL-1β factors as inflammatory indicators and the results It was investigated and analyzed with the one-way ANOVA test. result The results of this study showed that the expression levels of ABCA1 and IL-10 genes in the diabetic group significantly decreased compared to the control group, but the levels of IL-1β in the diabetic group showed a significant increase. The values of ABCA1 and IL-10 in the group receiving C-KIT+ showed a significant increase, but the values of IL-1β in the group receiving C-KIT+ showed a significant decrease compared to the diabetic group. Also, the values of ABCA1 and IL-10 genes in the diabetic group receiving C-KIT decreased and the amount of IL-1β increased in this group. Also, the histological study showed inflammatory changes in the left ventricle.
