Evaluation of relationship between CD34+ cells count and serum level of IL-1β, IL-6 , IL-18 and TNF-α before and after receiving G-CSF in Hematopoietic stem cell transplantation candidate patients

چکیده
The aim of this study was to investigate the relationship between hematopoietic stem cell count and surface Serum proinflammatory cytokines in hematopoietic stem cell transplant patients. Material and methods : 20 subjects including 12 autologous transplant patients and 8 healthy donors were used. Before and after receiving G-CSF, CD34 + cell count was performed by flow cytometry. Serum levels of cytokines IL-1β, IL-6, IL-18 and TNF- α was measured by ELISA and Real Time PCR was used to evaluate cytokine gene expression. Results: The results showed that the CD34 + cell count increased significantly compared to before G-CSF injection, but the cytokines IL-1β, IL-6, IL-18 and TNF-α significantly decreased in the donor's serum after chemotherapy. Healthy in allogues and patients in autologous, whose gene expression also confirmed this result, serum levels of these proinflammatory cytokines showed a significant inverse relationship with CD34 + cell count.
