Evaluation of the effects of total hydroalcoholic extract of Tanacetum kotschyi and meloxicam on neuropathy caused by vincristin in mice

Abstract
Introduction: In the process of the formation of neuropathy caused by the administration of anticancer drugs, several mechanisms have been proposed, including the activation of the oxidative stress system, increased activity of glutamate receptors, and increased activity of serotonin receptors. Meloxicam has anti-inflammatory and antioxidant effects and Tanacetum kotschyi extract also has oxidative stress inhibitory effects.Goal: The aim of this study was to investigate the preventive effect of meloxicam and total hydroalcoholic extract of Tanacetum kotschyi plant on neuropathic pain caused by vincristine injection in male mice.Materials and methods: 90 male mice in a weight range of 25 to 35 grams were randomly divided into 10 equal groups and underwent Tanacetum kotschyi Extract and Meloxicam for 25 days. Different doses of Extract and Meloxicam were injected three days before injection of vincristine (1 mg/kg, IP ) at 4th day. Then injection of Extract and Meloxicam continued for 3 days. After that we have 3 days for rest. Finally we performed hot plate test. This 7 day cycle repeated 3 times for all groups. At day 25, blood samples were collected for MDA and TAC estimation.Result: The results indicate a significant increase (p < 0.05) in the reaction time to pain in animals at a dose of 50 mg/kg of Tanacetum kotschyi extract and doses of 0.5 and 2 mg/kg of meloxicam, and also we saw a better effect of low dose meloxicam (0.5 mg/kg) in preventing neuropathy on most days of the experiment. Conclusion: Certain doses of Tanacetum kotschyi extract (50 mg/kg) and meloxicam (0.5 and 2 mg/kg) can be effective in preventing pain caused by neuropathy following the use of vincristine. In order to obtain the exact mechanism of neuropathy prevention, we will need additional studies.
