چکیده
In recent years, cancer treatment based on the immune system has attracted attention as an alternative and low-risk method, in which the immune cells are directed to a series of molecules (DAMP) that are emitted from cancer cells. These molecules cause the activation of APCs following T cell activation which can contribute to cancer cell elimination. This study is to investigate the immunogenic effect of emitted exosomes from dormant 4T1 cells treated with cold plasma in an animal model. In this study, new antigens of dormant cells tried to introduce to the immune system and create an anti-cancer effect against cancer cells. For this purpose, the cells (dormant and also non-dormant) were tried to make immunogenic death by cold plasma following DAMP emission and immune system motivation. Our first purpose was to induce dormant 4T1 cells by mimicking hypoxia by adding cobalt chloride solution to cell media. In this study, Two methods were used to create dormant cells. a) Mimicking hypoxia by cobalt chloride b) inducing starving stress by reducing FBS of cell culture. The dormant cell group (treated with plasma and nanoparticles) was eliminated from further tests owing to failure in inducing dormant cells. Instead of being the cells in the G0 phase after Cobal Chloride treatment, the cancer cells spread in different phases of the cell cycle. Next, 4T1 cells (non-dormant cells group) were exposed to Cold plasma and began to emit exosomes. These exosomes were injected into mice carrying breast tumors and their effects were studied. Our research showed the emitted exosomes from the cells treated with cold plasma can increase tumor suppression when compared to the positive and negative control groups. As a result, the exosome of breast cancer cells released by cold plasma can be used as a treatment method for breast cancer treatment.
