Expression comparison of HDAC2 in patients with sudden deaf

Abstract
According to the WHO, nearly 360 million people worldwide suffer from sudden hearing loss. In the present study, we aimed to compare the expression of HDAC2 gene in PBMC (Peripheral blood mononuclear cells) of peripheral blood of people with SSNHL with the expression of the same factors in the blood of healthy controls by Quantitative Real-time PCR. It is hoped that a deeper understanding of the molecular pathways involved in the pathogenesis of diseases will be a bridge to find new therapeutic solutions. Materials and Methods: From patients with SSNHL whose disease has been confirmed by a specialist physician, with the patient's consent, a complete demographic information checklist was obtained from the patient, blood samples were sent by the relevant physician for molecular studies. Then the results of markers and beds were compared. The samples were then transported to a molecular laboratory and a special RNA extraction kit was used to extract RNA from the blood. Then, using the PolyA RT-PCR method, all mRNAs in the sample were converted to cDNA, and after designing the primers, the presence of target genes was first investigated using a PCR device, and then after confirmation using a real-time PCR device. The expression level of these genes was measured in tumor samples and its margins. Finally, the data obtained from measuring the expression of these two genes were normalized by measuring the expression level of a house keeping gene called GAPDH and then compared. Results: The difference in HDAC-2 expression between case and control groups was statistically significant and this expression change was less in the patient group than the control group.
