Association study of rs2275913 single nucleotide polymorphism in IL-17A gene with Systemic lupus erythematosus (SLE) in Iranian patients

Patients with systemic lupus erythematosus (SLE) have more CD4+ cells producing IL-17 in compared to healthy individuals that is related to the level of disease activity. The association of interleukin (IL)-17 gene polymorphisms and susceptibility to SLE are in controversy. Here, the possible role of IL-17A rs2275913 polymorphism with SLE and its association with clinicopathological features were examined. Material and methods. A total of 80 patients with SLE and 80 age-matched healthy controls were included. The G-197A gene polymorphism was studied in peripheral blood mononuclear cells of participants using ARMS-PCR. The clinical and demographic data and SLE manifestations were recorded. The correlation of the studied polymorphism with the clinical data was investigated. Results. In SLE cases, the frequency of rs2275913 A allele was higher than controls (22.2% vs. 14.40%, respectively; OR =1.70, 95% CI = 0.95-3.02, p = 0.07). Besides, the frequency of AA and AG genotypes was higher in the SLE cases compared to the control group (33.8% vs. 22.5%, respectively); however, they were not statistically significant (p = 0.102). There were significant differences between AA genotypes and positive anti-dsDNA antibodies (p = 0.049) and also between GA genotype and skin manifestation (p = 0.045) in the SLE group. Fifty-seven percent (n = 45) of patients with SLE had SLEDAI above 6 and had active disease. No more significant associations were observed for the clinical and demographic data and SLE manifestations (p > 0.05).
