Impact of conditioned medium fibroblast cells on conversion molecular and morphological properties of luminal A and B subgroups in breast cancer

Heterogeneity of breast cancer cells is one of the problematic factors in the process of diagnosis and treatment of this disease. On the other hand, the influence of stromal factors in tumor tissue causes changes in the process of tumor progression, which ultimately challenges the treatment process. Fibroblasts, as the main and most abundant component of tumor microenvironment, play an important role in the interactions between cancer cells and microenvironment. Cancer-associated fibroblasts in angiogenesis and the production of growth factors that trigger immunosuppressive reactions. The transformation of different types of breast cancer into prognosis and different molecular and morphological features due to the activity of active fibroblasts in the microenvironment is important. Methods and materials: In this study, two subgroups of Luminal A and Luminal B of breast cancer were investigated. For this purpose, cell species MCF-7 and MDA-MB-361, which are types A and B, respectively, were prepared from Anistive Pasteur. HFFF-2 cells, which were cancer-associated fibroblasts, were used to activate three-dimensional culture (LOT) and medium fibroblast and spheroidal conditions were prepared. The studied cells were co-cultured with medium FBS10% and medium fibroblast and spheroidal condition and were studied after 24 and 48 hours. To study the morphology of cells and the possibility of their conversion to each other, after preparing light microscopic images, the amount of shape factor of two categories was calculated. Ki67 is a cell growth and proliferation protein that was calculated in our study to measure cell proliferation by immunocystochemistry with immunofluorescenty. Results: In this study, the biological and molecular effect of microenvironment on the possibility of conversion of subtypes of type A and B breast cancer in the same vessel with microenvironment in the treatment of breast cancer. According to the findings of this study, paracrine secretions of fibroblasts, both as spheroids and as fibroblasts, reduce the sensitivity to chemotherapy, which was determined by MTT and IC50 determination. Also, activated fibroblasts have shown a double effect in the study of cancer cell morphology in order to transform from epithelial to mesenchymal in both categories. In the study of Ki67 protein expression, the cell proliferation marker, which is more pronounced in Luminal A type, tends to convert the two subtypes into each other to prove it.
