Expression comparison of microRNA procecing elements (DROSHA, DICER, DGR8 and AGO) in patients with sudden deaf
Abstract
The aim of this study was to express the expression of genes involved in the microRNA maturation pathway that have been exposed to sudden deafness.
Materials and Methods: In this study, which is a case study. The sample size in each of the study groups (control-patient) was 40 samples, so that in the patient group 40 people with SSNHL whose disease has been confirmed by a specialist and in the control group 40 healthy people without hearing impairment who They were biologically identical to the patient group and were examined. After obtaining written consent from the participants (5 ml of blood was obtained). The PBMCs were first isolated and then rapidly transferred to -80 ° C. After collecting the samples, RNA was extracted using tresol solution; Then, cDNA synthesis for the genes was performed using special kDNA synthesis kits. Finally, using Real Time PCR technique and using specific primers for the desired genes, we evaluated the expression of target genes in the samples. To normalize the data in this study, we used the expression of GAPDH gene.
Results: In this study, the mean and standard deviation of age between the patient and healthy groups were 55.48 (2 2.3) and 56.80 (6 2.6) years, respectively. In the group of patients with SSNHL, the side of auditory involvement was left in 18 cases (0.45%) and right in 22 cases (55.0%), respectively. According to the severity of hearing loss in the patient group, the patient group was divided into 4 groups: mild, moderate, severe and deep. Each group had 10 cases (0.25%), 7, 18 cases (0.45%), and 5 cases, respectively 17%) and 5 cases (12.5%). The results of gene expression showed a significant decrease in DICER and DGCR genes in SSNHL patients compared to the control group, while the expression of the other two genes was not in a significant range despite changes in the two groups.
