Inhibition effect of HSP90 on TLR2 and TLR4 expression via MAPK pathway in Mellanoma cell lines (A-375) and (C6181)
Abstract
The aim of this study was to determine the effect of HSP90 on TLR2 and TLR4 in melanoma cell line (A-375).
Materials and Methods: The present in vitro study was performed on melanoma cancer cells in RPMI640 culture medium containing 10% FCS at 37 ° C and 5% CO2. After counting and dividing the cells, the cells inside the plate were stimulated in a time course with HSP90 protein. Incubation was performed. After 48 hours, the cells were collected and total mRNA was extracted and the expression levels of TLR2 and TLR4 were examined by qRT-PCR. Results: The results of the present study showed that the expression of TLR2 and TLR4 IL6 decreased in melanoma cell line (A-375) in exposure to HSP90. The results also showed the expression of NF-KB in melanoma cell line
