Expression comparison of let7 and DICER in tumoral and marginal tissues of tongue SCC specimens

Abstract
SCC of the tongue is the most common malignancy in the head and neck and can be treated with surgery. In terms of gender, the frequency of oral SCCs varies according to the age of the patients, so that in the elderly, men make up about 70% of cases, and in contrast, under the age of 45, the frequency in males is between 50 It is 65%. The aim of this study was to investigate the changes in the expression of DICER and let-7 genes in tumor tissue and marginal SCC of the tongue. Materials and Methods: In this cross-sectional laboratory study, people with SCC of the tongue undergoing surgery, and their cancer was confirmed by a pathologist, with the patient's consent, a complete demographic information checklist of Taken from the patient, samples from the tumor biopsy piece and tumor margin (0.5 to 30 mg) were sent by the relevant surgeon for molecular studies. A sample of the operated tumors was also sent to the pathology laboratory as usual. The number of samples studied is 20 tumors and its peripheral tissue (40 samples in total). Results: Comparison of DICER gene expression in tumor segment and tumor margin showed that the expression of DICER gene in tumor tissue was significantly higher than tumor margin tissue (p <0.01). Comparison of Let-7 gene expression in tumor segment and tumor margin According to the results, it can be seen that Let-7 gene expression in tumor margin tissue is significantly higher than tumor tissue (p <0.0001). There is a statistically significant relationship between DICER gene expression and tumor up regulation (p = 0.0049). Also, the expression level of Let-7 gene has a statistically significant relationship with down regulation, differentiation and stage of the tumor (p <0.0001, p = 0.0019 and p = 0.036, respectively).
