Abstract
Introduction: The ultimate goal of root canal therapy is to create a three-dimensional filling and impermeable seal throughout the length of the root canal and to properly match the filling material with the dentin wall to prevent the penetration of microorganisms and their products into the pericardial tissues. Lack of compliance with the walls of the canal or it’s loss for various reasons leads to periodontics complications. The goal of this study is to compare bacterial leakage in canals obturated with injected Gutta percha and AH26 sealer and canals obturated with single cone technique and Endoseal MTA . Methods and Materials: in this study the number of 50 mature human teeth with one canal were extracted, cleaned , and disinfected. The canals were cleaned and shaped .After preparing and shaping the canals with rotary file, the samples were randomly divided into 4 groups(Two experimental groups of twenty and two negative and positive control groups of five). The first experimental group was filled with injected Gutta Percha and AH26 sealer and the second experimental group was filled with single cone technique and Endoseal MTA.In the negative control group ,the canal were filled as in the first group and the positive control group was left without filling. In all specimens except the negative control group, all root surfaces except the apical two millimeters were sealed with two layers of cyanoacrylate glue. The samples then were tested for bacterial microleakage and microleakage days of up to 60 days were recorded. The data obtained from this study were analyzed by descriptive statistical methods and by reporting mean and standard deviation.Fishers exact test and Chi-square test was used to compare the micro-leakage in the test groups. A significant level in this study was calculated and resulted in p≤0.05 and statistical analysis was performed using SPSS 26 statistical software. Results: the number of samples with microleakage in single cone technique with Endoseal MTA was more in all days than injected gutta-percha and AH26.On the days 1to 35, this difference was not statistically significant.(P-value>0.05) On the day 40,the amount of microleakage in the single cone group was significantly higher than injected gutta-percha group.(P-value=0.025).On the days 45 to 60 ,the number of samples with microleakage in the single cone group was more than injected gutta-percha group, but this difference was not statistically significant.(P-value>0.05) Conclusion:The results of the present study showed that the number of samples with microleakage in single cone technique and Endoseal MTA was more in all days than injected gutta-percha and AH26 group. However, based on exact Fishers test and Chi-square test, this difference was not statistically significant.
