Expression comparison of microRNA-221 and microRNA-143-3p in patients with TBI

Abstract
Multiple brain injuries due to accidents and work accidents are one of the leading causes of death in the country. Despite the clinical significance of these injuries, no individual has been offered to accurately identify and estimate traumatic injuries. MicroRNAs are regulatory molecules with a length of 19 to 25 nucleotides and are the product of shRNA or pre-miR. The aim of this study was to investigate the changes in some microRNAs in patients with spinal cord injuries and their role in the prognosis of patients. Materials and Methods: In this study, which is a case study. The sample size in each of the studied groups (control-patient) was 100 samples, so that in the patient group 100 patients with head trauma whose disease has been confirmed by a specialist and in the control group 100 healthy individuals without nervous system injuries in terms of Biologically matched to the patient group were examined. After obtaining written consent from the participants (5 ml of blood was obtained), the sera were first separated and then immediately transferred to -80 ° C. After collecting the samples, RNA was extracted using tresol solution; Then, cDNA synthesis for genes was performed using special kDNA synthesis kits. Finally, using Real Time PCR technique and using specific primers for the desired genes. Results: In this study, the mean and standard deviation of age between patients and healthy groups were 35.26 (12 12.1) and 34.92 (6 12.6) years, respectively. In the group of patients with TBI, in 18 cases (0.18%) were female and in 82 cases (88.0%) were male. The results of the gene expression study showed a significant increase in the studied genes in patients with TBI compared to the control group; However, ROC curve analyzes have shown that both genes can act as biomarkers in identifying people with trauma.
