Surface plasmon resonance signal enhancement by means of drug loaded magnetic nanoparticles for interaction of erlotinib with lung cancer cells

At the present study, magnetic nanoparticles (MNPs) were used as signal enhancement tool for evaluation of erlotinib-loaded magnetic nanoparticles interaction with human lung cancer cells compared to erlotinib alone. Material and methods: Human lung cancer cells (A549 cell line) were cultured on SPR Au sensor chip. Different concentrations of Erlotinib and MNP-erlotinib solutions (5,10,20,40,80 µg/ml) were prepared and injected to channel 1 and 2, respectively. The interaction of erlotinib with A549 cells was studied with and without conjugation of MNPs at three temperatures (20,25,37°c). Finally, signal enhancement and kinetic parameters were evaluated. Results: Signal enhancement evaluation resulted in about 2.5-fold increase in signal height at MNP-erlotinib interaction with A549 cells. Furthermore, the results of kinetic study indicated that erlotinib and MNP-erlotinib affinity toward A549 cells was increased as temperature rose from 20 to 37°c.
