The effect of hydatid cyst fluid on the expression of microRNA-1 and the growth of cancer cells MCF-7 and MDA-MB-231

Abstract
The aim of this study was to evaluate the effect of hydatid cyst fluid on the growth of breast cancer cell lines including MDA-MB-231 and MCF-7 and changes in the expression of miR-1 in cell lines untreated and treated with hydatid cyst fluid in vitro. Matherial and methods: Two breast cancer cell lines including MDA-MB-231 and MCF-7 were cultured in culture media thenandand MTT assay was performed to determine IC50 of hydatid cyst fluid.Cells were treatedby 18.5 μM hydatid cyst fluid. Then RNA was extracted and cDNA was synthesized. Finally, changes in the expression of miR-1 using QRT-PCR in two groups untreated and treated breast cancer cell lines with hydatid cyst fluid was determined. Result: MTT results showed that growth and proliferation of MDA-MB-231 and MCF-7 cell lines decreased that showed a cytotoxic effect of hydatid cyst fluid on this breast cancer cell lines. QRT-PCR also showed that miR-1 expression level was increased in MDA-MB-231 (P< 0.0203) and MCF-7 (P< 0.0394) cell lines treated with hydatid cyst fluid.
