Investigating the solubility of celecoxib through cosolvency method

Abstract
Solubility is one of the most significant physicochemical properties of drugs and improving the solubility of drugs is still a challenging subject in pharmaceutical sciences. The solubility of a drug substantially depends on the physical and chemical properties of the solute and solvent as well as on temperature. Celecoxib, according to Bio-pharmaceutical Classification Systems is a type 2 drug. Increasing the solubility of this group can lead to increased bioavailability. Aims: Solubility determination of celecoxib in binary mixtures of 3 different solvents, including water, 2-propanol (2-P), and 1-Methyl-2-pyrrolidone (NMP), at different temperature and checking the predictability of some previously developed mathematical methods, are the aims of this work. Methods and Materials: The solubility of celecoxib in binary mixtures was determined using the saturating shake-flask method of Higuchi and Connors. Shaker was placed in an incubator due to setting the desired temperature. Result and Discussion: The solubility data of celecoxib in binary solvent mixtures at different temperatures indicate that the solubility of celecoxib was improved dramatically due to presence of cosolvent in comparison with the solubility of celecoxib in water solely, and this improvement also depends on increasing temperature. The recorded data in different temperature states that the maximum solubility of celecoxib in binary mixture is in neat cosolvent. Among NMP and 2-P, NMP is a better solvent for celecoxib. Conclusion: The solubility profile of celecoxib in aqueous solvent mixtures and non-aqueous solvent mixtures, show a nonlinear pattern with maximum solubility in neat non-aqueous cosolvents. Cosolvency method with increasing temperature leads to a significance solubility development of celecoxib. Different cosolvency models, were applied to correlate solubility data of celecoxib in binary solvent mixtures.
