Investigation of hsa-miR-101 and hsa- miR-200a genes expression during osteoblastic differentiation of human mesenchymal stem cells
Abstract
The purpose of this study was to recognize the pattern of miRNA-101a-3p and miRNA-200a expression during osteoblastic differentiation of human adipose tissue-derived mesenchymal stem cells (hADSC).
Methods: The cells were incubated in osteoblastic differentiation medium for a period of 21 days. To confirm the successful differentiation cells into osteoblast cells, Alizarin Red S staining was performed. The expression level of miRNA-101a-3p and miRNA-200a were analyzed by real-time PCR during 0, 7, 14 and 21 days during differentiation induction.
Results: Data exhibited the increase of extracellular red color deposition which was evident at the end of the incubation period. Based on our data, the expression of miRNA-101a-3p and miRNA-200a was induced which indicated with uptrend pattern. miRNA-101a-3p showed a significant increase (P <0.05) about 4.1 fold on day 21 compared with the control sample, and miRNA-200a on day 21 increased significantly (P <0.05) to 2.5 fold compared to control sample.
