Study of the anti-angiogenic response of endothelial cells to the treatment with Docetaxel anti-cancer agent
Abstract
The aim of the present study was to evulate the effect of doctaxel on the expression level of angiogenic factors on endothelial cells
Methods: In this study, HUVEC cell line of gastric adenocarcinoma cells were cultured in RPMI1640 medium, then determined by MTTassay of effective dose and after 24 hours treatment with and Docetaxel changes of gene expression. The study was compared to the control group by Real Time PCR method. To evaluate the expression of the studied genes(mir106b,mir-21,mir-155 and HIF-1@). Results: Treatment of cells with DOC induced changes in expression of the studied genes. Based on the results of the study,the expression of metastasis-dependent genes was significantly increased in the treated cells compared to the untreated cells. The expression of MicroRNAs also significantly decreased in DOC-treated cells. In other words, DOC resulted in altered expression of genes involved in metastasis and angiogenesis.
