Inhibition effect of HSP90 on HMGA1,HMGB1 via NF-KB and MAPK in melanoma cell line
Abstract
Melanoma is the most invasive and deadly type of skin cancer and one of the malignant cancers. Various methods such as radiotherapy, chemotherapy, immunotherapy and surgery are used to treat this type of cancer. The aim of this study was to determine the effect of HSP90 on HMGA1, HMGB2 in melanoma cell line (A-375).
Materials and Methods: The present in vitro study was performed on melanoma cancer cells in RPMI640 culture medium containing 10% FCS at 37 ° C and 5% CO2. After counting and dividing the cells, the cells inside the plate were stimulated in a time course with HSP90 protein.. The growth and proliferation of cancer cells were assessed by MTT technique. The collected data were analyzed using Student t-test and SPSS software and P <0.05 was considered significant in all statistical tests.
Results: HSP90 expression was significantly increased in melanoma cell line. Expression of HMGA1 and HMGB2 was significantly decreased by HSP90 in A-375 cell line. MAPK and NF-KB levels in A-375 cell line decreased with HSP90 stimulation.
