MiR-143 replacement effect on growth and migration inhibition in lung cancer cell line (A549)
Abstract
The aim of this study was to investigate the change of expression level of miR-143 and target genes of this microRNA after replacement in the lung cancer cell line (A549) and its effect on inhibiting the growth and migration of these cells.
Material and methods: IC50 of Antibiotic Geneticin (G418) was obtained in the A549 cell line using the MTT assay. miR-143 was transfected into lung cancer cell lines (A549) by using jetPEI. qRT-PCR test was used to demonstrate the increased expression level of the miR-143 and targeted genes. MTT test was used to show inhibition of proliferation of lung cancer cells. Wound healing assay was used to examine the status of transfected cancer cell migration.
Results: By using MTT test, the IC50 of Geneticin antibiotic was obtained 600 µg/ml. The result of the qRT-PCR test showed the increased expression level of miR-143 and decrease targeted genes expression in A549 transfected cells with miR-143. The MTT test results demonstrate inhibition of cancer cells proliferation. Wound healing assay results showed migration reduction of transfected cells with miR-143 compared to negative control cells.
