Determination of phenyl-butyrate effects on sensibility increasing of oxaliplatin at colon cancer rat animal model

Abstract
Introduction: Colorectal cancer is the 3rd most common type of cancer worldwide. The Oxaliplatin drug, which is commonly used in chemotherapy regimen of colorectal cancers, acts inspecifcally and cause many side effects. The previous studies have shown that intestinal microbial fermentation products such as Butyrate have broad antitumor effects through inhibition of histone deacetylase enzyme and induction of apoptosis. Purpose: The purpose of this study was to investigate the effect of Phenylbutyrate on Oxaliplatin anticancer effect in colorectal induced wistar Rat model. Procedure: A total of 30 male wistar Rat, were divided into 5 groups of 6 Rats. One group considered as normal control group, and the rest of them were subjected to colorectal cancer induction though 24 intraperitoneal injection of the Dimethylhydrazine. The cancerous rats were divided into the following groups: “Cancerous without treatment”; “Oxaliplatin treated”; “PhenylButyratetreated”; and “Oxaliplatin-PhenylButyrate combination treated”. The rats were treated for four weeks. Then, the rats were underwent surgery and their colon was monitored regarding tumors number and size. Findings: There is 32% reduction in number and 50% decrerase in size of tumors in “Oxaliplatin treated” group in coparesion to “Cancerous without treatment” group. Moreover, the results of the Phenylbutyrate treatment indicated that the “Phenyl Butyrate treated” group has 75% and 35% reduction in number and size of tumors respectively. At last, in comparision of “Oxaliplatin-PhenylButyrate combination treated” with “Cancerous without treatment” groups, there is 75% and 66% reduction in number and size of tumors respectively. Conclusion: It can be concluded that overally Oxaliplatin has great influence in the reduction of tumors size and Phenylbutyrate has great impact on tumors number. Furthermore, the results indicated that the combination therapy of Oxaliplatin and Phenylbutyrate has higer efficacy in comparesion with each drugs alone.
